Diabete sotto controllo con il ‘rubinetto molecolare’

Scienziati californiani hanno scoperto un ”rubinetto molecolare” chiudendo il quale si può ridurre la produzione di zucchero da parte del fegato che nei pazienti con la forma più diffusa di diabete, il diabete insulino-resistente, è eccessiva. Gli esperti, in un esperimento su topolini diabetici i cui risultati sono stati resi noti sulla rivista Nature, hanno dimostrato che chiudendo questo ‘rubinetto’ con specifici ‘tappi molecolari’, il fegato riduce la produzione di zucchero e quindi la glicemia può essere tenuta sotto controllo. La ricerca è stata condotta da Marc Montminy del Salk Institute for Biological Studies di La Jolla (California). Nel diabete insulino-resistente l’organismo è sordo ai messaggi dell’insulina di produrre meno zucchero e incamerare quello già sufficiente, quando non eccessivo, arrivato coi pasti. In più, nei diabetici l’ormone del digiuno – il glucacone – che si attiva quando digiuniamo e la glicemia si abbassa, lavora troppo. Questo porta all’iperattivazione di un altro ormone, il ‘CRTC2’, che attiva a sua volta l’IP3, il ‘rubinetto’ molecolare, che tecnicamente è un canale sulla superficie delle cellule del fegato. Quest’ultimo, una volta aperto, scatena una cascata di reazioni che innesca la produzione di zucchero da parte del fegato. Zucchero poi riversato in eccesso nel sangue. Così la glicemia sale. La ricerca del Salk Instituto è ancora di base, ma se la strada fosse percorribile nell’uomo potrebbe portare a una svolta nella lotta al diabete che è ormai una piaga dei Paesi ricchi e ipernutriti.
